5' capping of respiratory syncytial virus (RSV) mRNAs is catalyzed by the polyribonucleotidyl transferase and methyltransferase domains of the catalytic subunit L of the RNA-dependent RNA polymerase (RdRP) complex. The capping of RSV mRNAs is a two-step process, involving cap0 and then cap1 creation. The so-called cap0, labeled m7GpppG- as all RSV transcripts start with guanidine (Liuzzi et al. 2005), is created through N7 methylation catalyzed by the methyltransferase domain of the L protein. The complex of L and P proteins is sufficient for the N7-methylation to occur. In RSV, N7-methylation (cap0 creation) precedes 2'-O-methylation (cap1 creation) (Sutto-Ortiz et al. 2021). 5' capping was confirmed in: 1C (NS1) mRNA (Collins and Wertz 1985; Liuzzi et al. 2005), 1B (NS2) mRNA (Collins and Wertz 1985; Liuzzi et al. 2005), N mRNA (Liuzzi et al. 2005), P mRNA (Liuzzi et al. 2005), M mRNA (Satake and Venkatesan 1984; Liuzzi et al. 2005), SH mRNA (Liuzzi et al. 2005), G mRNA (Liuzzi et al. 2005), F mRNA (Elango et al. 1985; Liuzzi et al. 2005), M2 mRNA (Liuzzi et al. 2005), and L mRNA (Liuzzi et al. 2005).
