Cry and Per proteins form complexes with the kinase Csnk1d (Ck1delta) and/or Csnk1e (Ck1epsilon) and bind the Bmal1:Clock heterodimer at promoters of target genes. The Cry:Per:kinase complex thereby brings Csnk1d and/or Csnk1e into proximity with Clock, resulting in the phosphorylation of Clock (Cao et al. 2021). The phosphorylation may occur at serine-38 and serine-42 in the DNA-binding domain of Clock (Yoshitane et al. 2009), as the phosphorylation of Clock by Csnk1d causes repression of Bmal1:Clock transactivation activity and dissociation of the Bmal1:Clock heterodimer from DNA (Cao et al. 2021).
