Cryptochrome (CRY) and Period (PER) proteins form complexes with the kinase CSNK1D (CK1delta) and/or CSNK1E (CK1epsilon) and bind the BMAL1:CLOCK heterodimer at promoters of target genes. The CRY:PER:kinase complex thereby brings CSNK1D and/or CSNK1E into proximity with CLOCK, resulting in the phosphorylation of CLOCK (inferred from mouse homologs).The residues phosphorylated by CSNK1D,E may be serine-38 and serine-42, which are located in the DNA-binding region of CLOCK and are observed to undergo cyclic phosphorylation that causes weakened transactivation and DNA-binding activity of BMAL1:CLOCK (inferred from mouse homologs in Yoshitane et al. 2009).
