STAT1 and NFATC1 bind the conserved noncoding region 12 (CSR 12) of the TBX21 (T-bet) gene (inferred from mouse homologs in Zhong et al. 2025). TBX21 expression is initiated by IFNG signaling via STAT1 (inferred from mouse homologs in Lighvani et al. 2001, Afkarian et al. 2002) phosphorylated on serine 727 (inferred from the mouse homolog in Maldonado et al. 2009) and by TCR signaling via STIM1, calcium signaling, and NFATC1 (inferred from mouse homologs in Zhong et al. 2025). TBX21 then activates expression of the IL21 receptor subunit IL21RB2, allowing maintenance of TBX21 expression by IL21 acting via IL21RB2 and STAT4.
