ITGA2 encodes integrin alpha-2, an integrin receptor that dimerizes with ITGB1 (integrin beta-1) to bind laminin, collagen, fibronectin and E-cadherin in the extracellular matrix (Emsley et al, 2000; Emsley et al, 2004; Tiger et al, 2001; reviewed in Campbell and Humphries, 2011; Pang et al, 2023). MITF-M binds to the ITGA2 gene to repress its expression in melanoma cells as assessed by MITF-M knockdown, ChIP-seq, microarray and CUT-and-RUN analysis (Dilshat et al, 2021; Webster et al, 2014; Laurette et al, 2015).
MITF-M represses the expression of genes involved in extracellular matrix organization, focal adhesions and epithelial-to-mesenchymal transitions to impact the invasive/proliferative phenotype of the cell. Low levels of MITF-M are associated with a quiescent/invasive phenotype, while higher levels of MITF-M support proliferative growth (Dilshat et al, 2021; Carreira et al, 2006; Hoek et al, 2008; reviewed in Rambow et al, 2019).
