SERPINE1, also known as Plasminogen activator inhibitor I (PAI-1), is a serine protease inhibitor with canonical roles in fibrinolysis (reviewed in Morrow and Mutch, 2023). More recently, roles for SERPINE1 have been identified in extracellular matrix remodeling, tumor cell invasion and migration independent of its protease activity (Kjoeller et al, 1997; Degryse et al, 2004; reviewed in Pavon et al, 2016). SERPINE1 inhibits the formation of plasmin at the plasma membrane, restricting the plasmin-dependent degradation of the ECM (reviewed in Pavon et al, 2016).
SERPINE1 has been identified as a direct target of MITF-M-dependent regulation in melanoma cells by ChIP-seq, CUT-and-RUN DNA binding analysis and CRISPR- and si/shRNA-mediated MITF-M knockdown (Dilshat et al, 2021; Webster et al, 2014; Laurette et al, 2015).
