POLRMT initiates transcription at a stem-loop structure in OriL and polymerizes a primer RNA of approximately 25 nt (Wanrooij et al. 2008, Fusté et al. 2010, Sarfallah et al. 2021, inferred in vivo from mice in Wanrooij et al. 2008). The transcription factor TFAM is not required for transcription initiation at OriL (Fusté et al. 2010). Slippage of the transcript at 6 deoxythymidine residues results in an extra 2 adenosine residues being incorporated in the RNA primer and this slippage is required for stability of the initiation complex (Sarfallah et al. 2021)
