WNT signaling activates expression of Eomesodermin (EOMES), which in turn activates expression of SOX17 in precursors of primordial germ cells (PGCs), possibly by binding the SOX17 gene (Chen et al. 2017, Kojima et al. 2017). BMP4 signaling, required for specification of PGCs, enhances expression of SOX17 in precursors of PGCs (Kojima et al. 2017). SOX17 is a critical factor for the differentiation of both PGCs and endoderm and it appears to act combinatorially with other factors to generate either PGCs or endoderm. PRDM1 represses genes expressed in endoderm and hence facilitates the formation of PGCs (Irie et al. 2015).
