AKT3-phosphorylated TBX3 binds to the promoter of ID1 to activate expression, as assessed by electrophoretic mobility shift assays, chromatin immunoprecipitation and reporter gene assays (Peres et al, 2021). ID1 is a member of the ID1 - 4 family of helix-loop-helix proteins that lack the DNA binding region and consequently act as dominant negative inhibitors of other HLH-containing proteins upon heterodimerization (Massari and Murre, 2000; reviewed in Perk et al, 2005; Ling et al, 2014). In osteoclast cells, ID1 has been shown to bind to MITF, and MITF has been shown to directly activate CDH1 expression (Lee et al, 2006; Mansky et al, 2002; Dilshat et al,2021). In melanoma cells, TBX3 also upregulates ID1 expression which prevents MITF from binding and activating the CDH1 promoter (Peres et al, 2021).
