Based on homology of the C-terminal lumenal domains of ATF6 and ATF6B, ATF6B is inferred to interact with HSPA5 (BiP, GRP78) and dissociate from HSPA5 as HSPA5 binds unfolded protein. The release of HSPA5 from ATF6B allows ATF6B to traffic from the endoplasmic reticulum to the Golgi where it is activated by cleavage.
