Colony-stimulating factor 1 receptor (CSF1R, CSF-1R) is a protein tyrosine kinase transmembrane receptor for colony-stimulating factor-1 (CSF1, macrophage CSF, M-CSF) and for interleukin-34 (IL-34) (Wang et al. 1993, Lin et al. 2008, Chihara et al. 2010, Ma et al. 2012, Felix et al. 2015). Responses of CSF1R to CSF1 and IL34 are qualitatively similar but quantitatively different (Chihara et al. 2010). CSF1R is a transmembrane glycoprotein and contains five extracellular ligand binding immunoglobulin-like domains, a single transmembrane segment, and a split intracellular kinase domain. CSF1 is a disulfide linked homodimer and either expressed as a transmembrane molecule or proteolytically processed to produce soluble glycoprotein or proteoglycan forms (Stanley and Chitu, 2014). IL-34 is a non-covalently linked homodimer that is secreted (Lin et al. 2008).
CSF1R binds with its extracellular D2 and D3 domains to CSF1 or IL-34. One CSF1 subunit binds to one CSF1R molecule, the second CSF1 subunit binds another CSF1R molecule, resulting in dimerization of CSF1R through the interaction of its D4 and D5 domains (Wang et al. 1993, Chihara et al. 2010, Ma et al. 2012, Felix et al. 2012).
