A distinct BAF complex has been identified in mouse and human embryonic stem cells (esBAF) (Ho, Jothi et al, 2009, Ho, Ronan et al, 2009; Zhang et al, 2014; reviewed in Kadoch and Crabtree, 2015; Cenik and Shilatifard, 2021; Innis and Cabot, 2020). esBAF is characterized by the presence of SMARCA4/BRG1 to the exclusion of SMARCA2/BRM and SMARCD1/BAF60A. Where mouse esBAF uses only SMARCC1 (BAF155), human esBAF appears to contain both SMARCC1 (BAF155) and SMARCC2 (BAF170) (Ho, Ronan et al, 2009; Ho, Jothi et al, 2009; Zhang et al, 2014; Wade et al, 2015). esBAF also contains subunits BCL11A/B (Kadoch et al, 2013; reviewed in Kadoch and Crabtree, 2015; Innis and Cabot, 2020; Cenik and Shilatifard, 2021). esBAF colocalizes with each of the pluripotency factors, and overexpression of esBAF subunits induces pluripotency in fibroblasts (Ho, Ronan et al, 2009; Ho et al, 2011). As the assembly of human esBAF has not been examined in detail, this pathway is based on the modular assembly of cBAF, pBAF and ncBAF (Mashtalir et al, 2018; Mashtalir et al, 2020). |
