SARS-CoV-2 infection down-regulates class I MHC in both infected human angiotensin?converting enzyme 2 (hACE2)-expressing HEK293T cells and infected lung epithelial cells of hACE2 transgene mice (Zhang Y et al. 2021).
SARS-CoV-2 ORF8 (8), which has no orthologous protein in SARS-CoV-1 (Li T et al. 2020), was found to regulate the expression of class I MHC on the surface of the infected cells through an autophagy-dependent lysosomal degradation of class I MHC. Knockdown of viral 8 protein expression in SARS-CoV-2?infected cells restored class I MHC expression (Zhang Y et al. 2021). This interaction leads to impaired antigen presentation.
