text The Tbxt (T, Brachyury) gene is transcribed to yield mRNA and the mRNA is translated to yield Tbxt protein. Expression of Tbxt is activated by Wnt signaling via beta-catenin (Ctnnb1) (Yamaguchi et al. 1999, Arnold et al. 2000, Morkel et al. 2003) and Bmp4. Tbxt is initially expressed in the primitive streak during gastrulation, then throughout the presumptive mesoderm, and then becomes restricted to notochord and tail bud (Herrmann 1991, Kispert and Herrmann 1994, Lolas et al. 2014). Expression is lost with maturation of the notochord. Expression of Tbxt and Eomes are required for exit from pluripotency and expression of mesoderm and definitive endoderm gene programs (Tosic et al. 2019). Oct4 has been suggested to be involved in mesoderm induction and regulation of Tbxt (Thomson et al. 2011)
