text CXXC5 expression is estrogen-responsive and depends on the interaction of ligand-bound ESR1 with an ERE at -242 of the promoter (Nott et al, 2009; Ya?ar et al, 2016). CXXC5 is a transcription factor that plays roles in cellular proliferation and differentiation (Andersson et al, 2009; Kim et al, 2010; Aras et al, 2013; Li et al, 2014; Kim et al, 2016; Kim et al, 2015; Lee et al, 2015; Wang et al, 2013) KANK1 is an additional positive regulator of CXX5 expression. KANK1 is a regulator of cytoskeleton rearrangements that has been identified as a putative tumor suppressor gene that promotes CXXC5-dependent apoptosis (Wang et al, 2006; Kakinuma et al, 2008; Roy et al, 2009; Li et al, 2011; Cui et al, 2017). KANK1 mutations have been identified in renal cell carinomas, malignant peripheral nerve sheath tumors (MPNST), and cancers of the prostate, pancreas, uterus and stomach (Sarkar et al, 2002; Medves et al, 2010; Cui et al, 2017). Re-expression of KANK1 in depleted MPNST cell lines and mouse xenograph models decreases cell growth rates, increases CXXC5 protein levels and increases CXXC5-mediated apoptosis (Cui et al, 2017).
