text The Ser/Thr protein kinase, Fused (FU), is capable of autophosphorylation and is proposed to be activated by phosphorylation on at least Thr158 and probably on other residues too. Costal2 (COS), bound to the phosphorylated transmembrane receptor Smoothened (SMO), is itself phosphorylated by FU at Ser572 and Ser931. Both FU and COS promote phosphorylation of SMO and its accumulation at the plasma membrane resulting in a positive feedback loop.
