A dimer of phosphorylated Bmal1:Clock (Arntl:Clock) heterodimers binds adjacent E-boxes (EE element, E1E2 element) in the promoter of the Per2 gene (Akashi et al. 2006, Nakahata et al. 2008, Mongrain et al. 2011, Rey et al. 2011, Koike et al. 2012) and recruit the histone acetyltransferase Crebbp (Koike et al. 2012) and the histone methyltransferase Kmt2a (Mll1) (Katada and Sassone-Corsi 2010) to activate transcription. Large scale chromatin immunoprecipitation also indicates that Bmal1:Clock binds additional sites in the Per2 promoter (Rey et al. 2011, Koike et al. 2012). Npas2 can heterodimerize with Bmal1 in place of Clock, and the Bmal1:Npas2 heterodimer binds E-boxes and regulates Per2 expression (Reick et al. 2001, Yoshii et al. 2015, Landgraf et al. 2016).
