Expression of Sox17 in definitive endoderm
Sox17 is first expressed in the visceral endoderm, then in presumptive definitive endoderm before and during intercalation with visceral endoderm (Viotti et al. 2014). Wnt signaling activates Sox17 expression through the binding of Lef1:Ctnnb1 to the Sox17 promoter (Engert et al. 2013). Foxa2 and Gata4 also directly activate Sox17 by binding a distal enhancer (Trinh et al. 2022). Mixl1 also has role in activating Sox17 and other endodermally expressed genes (Mohn et al. 2003, Izumi et al. 2007, Lim et al. 2009). Loss of Mixl1 results in reduced recruitment of definitive endoderm and defective migration of endoderm during gastrulation (Hart et al. 2002).
