Notch1 binds Dlk1
Using a yeast two-hybrid system, mouse Notch1 and Dlk1 were found to interact and this interaction involves EGF repeats of both proteins. Co-expression of Notch1 and Dlk1 in cis results in inhibition of Notch1-mediated transcription. Notch1 and Dlk1 levels increase at confluence in mouse fibroblast cell culture, which is accompanied by a decreased transcription of the Notch1 target Hes1. Therefore, Dlk1 is implicated as a negative regulator of Notch1 signaling in cis.
