TP53 binds to the ARG1 genomic region to repress transcription as assessed by ChIP and reporter gene assay (Li et al, 2019). TP53-mediated downregulation of urea cycle genes contributes to cellular proliferation by increasing the levels of polyamines like putrescine through an ammonia-dependent upregulation of ornithine decarboxylase (ODC) translation, the key enzyme in polyamine synthesis (Li et al, 2019; reviewed in Holbert et al, 2022).
