Host 5'-3' exoribonuclease 1 (XRN1) catalyzes the hydrolysis of DENV decapped mRNA from 5' to 3', yielding ribonucleotide-5'-monophosphates. The degradation stops at discrete elements of the DENV RNA 3′UTR that confer resistance to XRN1 (so called Xrn1-resistant RNA aka xrRNA) (Chapman et al., 2014a,b; MacFadden et al., 2018). The residual product of this incomplete degradation is referred to as small or subgenomic flaviviral RNA (sfRNA). Dengue viruses generate three to four distinct species of sfRNAs. They inhibit host antiviral responses by interacting with viral and host proteins (reviewed by Slonchak & Khromykh, 2018; Liu et al., 2023). sfRNAs appear to function as reversible inhibitors of XRN1 (Moon et al., 2012; Chapman et al., 2014a).
