The nucleocapsid complex, localized on the cytosolic side of the ER membrane, associates with the viral glycoproteins on the lumenal side of the ER membrane, and buds into the ER lumen, forming an immature virion (reviewed by Apte-Sengupta et al., 2014; Barnard et al., 2021). Budding can also happen without capsids (e.g., formation of subviral particles), indicating that the mechanisms ensuring efficient nucleocapsid incorporation into budding viral membranes are not well understood (reviewed by Apte-Sengupta et al., 2014). The lipid content of DENV-2 virions differs significantly when produced in mammalian (rhesus monkey kidney LLC-MK2) versus insect (Aedes albopictus C6/36) cells (Hitakarun et al., 2022). As placeholder for all included lipids we show phosphatidylserine (PS). Formation of infectious DENV-2 virions and their subsequent secretion depends on Y-box-binding protein 1 (YBX1) (Diosa-Toro et al., 2022, using DENV-2 NGC), as well as nucleolin (NCL; Balinsky et al., 2013, using DENV-2 Tonga/74; reviewed in Barnard et al., 2021). Tumor susceptibility gene 101 (TSG101) protein, part of the Endosomal Sorting Complexes required for Transport (ESCRT), is required for viral budding (Tabata et al., 2016, using DENV-2 NGC).
