Dengue virus proteins NS2B and NS3 bind non-covalently. The NS2B:NS3 protease complex (aka NS2B3) is essential in both the viral multiplication cycle and is implicated in disease pathogenesis (Falgout et al., 1991; Noble et al., 2012; reviewed in Nitsche et al., 2014; Leonel et al., 2018). While anchored to membranes, the catalytic domain of the NS2B3 complex, including the hydrophilic region of NS2B that acts as a cofactor, is oriented towards the cytosol where it encounters and processes its substrates (see, e.g., Wahaab et al., 2021). DENV-1 NS2B3, depending on the state of replication, can localize to the nucleus, however (Gandhi et al., 2023).
