An unknown endoplasmic reticulum (ER)-membrane-bound host peptidase cleaves the DENV-4 NS1-NS2A junction, producing NS1 and NS2A; this mechanism was supported by experiments using monkey CV-1 cells, human TK-143 cells, and in vitro assays with dog pancreas microsomes (Falgout et al., 1995; reviewed in Verhaegen & Vermeire, 2024). We are inferring the same process with DENV-2 proteins from these results.
