Based on findings in Drosophila (Cordle et al. 2008) and in mammalian cell culture where human NOTCH1 and rat Dll1 were used (Sprinzak et al. 2010), binding of NOTCH2 to DLL/JAG ligands expressed in the same cells (in cis) is expected to block NOTCH2 activation by DLL/JAG ligands expressed on neighboring cells (in trans). Cis-inhibiton of NOTCH signaling can amplify small differences in NOTCH and DLL/JAG levels between neighboring cells.
