The tripartite efflux system acrA:acrB:tolC is the predominant efflux system in members of enterobacteria. It has a wide substrate profile, consisting of multiple different classes of antibiotics (chloramphenicols, tetracyclines, quinolones, macrolides, and β-lactams) and other xenobiotics like n-alkane solvents (Tsukagoshi & Aono, 2000). acrB is a transport protein of the Resistance-Nodulation-Cell Division (RND) superfamily which localizes to the plasma membrane. Together with the acrA bridge acrB is bound to the tolC trimer porin in the outer membrane, In the second reaction catalyzed by the tolC trimer, substrate diffuses to extracellular space (Zgurskaja & Nikaido 1999; reviewed by Alav et al., 2021).
