PTPN1 (PTP1B) interacts with Annexin A2 (Ye et al. 2021) and dephosphorylates tyrosine-24 of Annexin A2 (ANXA2) (Zhang et al. 2020) in response to turbulent flow across endothelial cells. Serine-26 of ANXA2 remains phosphorylated (Zhang et al. 2020). Activation of the mechanosensitive cation channel PIEZO1 is required for dephosphorylation of ANXA2 (Zhang et al. 2020). PTPN1 may be activated by calcium-dependent cleavage by calpain (Frangioni et al. 1993, de Jong et al. 2014).
