MITF binds to promoter elements in the upstream regulatory region of the DICER1 gene to activate it expression as assessed by ChIP, reporter gene assay and qRT-PCR (Levy et al, 2010).
The DICER1 gene encodes the RNase III endonuclease that plays a key role in generation of mature microRNAs for post-transcriptional gene silencing. DICER1 expression is essential for melanocyte survival in vitro and in vivo (Levy et al, 2010). MITF-mediated expression of DICER1 may contribute to melanocyte survival through its processing of the miR17~92 cluster, which targets the pro-apoptotic factor BIM1 (also known as BCL2L11) (Levy et al, 2010; Yamamura et al, 1996; Trisciuoglio et al, 2005; Fontana et al, 2008; Koralov et al, 2008; Ventura et al, 2008; Xiao et al, 2008; reviewed in Mendell et al, 2008; Goding and Arnheiter, 2019).
