AKT1 is phosphorylated in osteocytes in response to mechanical loading and directly phosphorylates Connexin43 (Cx43, GJA1) on serine-373 and integrin alpha5 (ITGA5) on an unknown residue (inferred from mouse homologs). Phosphorylation of the hexameric GJA1 hemichannel enhances its interaction with ITGA5 and also enhances the opening of its pore, resulting in an efflux of prostaglandin E2 (PGE2) and ATP (inferred from mouse homologs).
