PIEZO1 is a non-selective cation channel that passively transports Ca2+, K+, Mg2+, and Na+ ions across the plasma membrane in response to mechanical force applied to the membrane (Albuisson et al. 2013, Andolfo et al. 2013, and inferred from the mouse homolog). In resting, polarized cells, opening of PIEZO1 channels initiates an influx of Ca2+ and Na+ ions and an efflux of K+ ions. PIEZO1 forms trimers with a central channel and three blades that protrude in the plane of the membrane (inferred from the mouse homolog). The PIEZO1 trimer is not flat in the plane of the membrane, but rather forms a dome (inferred from the mouse homolog). Application of force to the membrane opens the central channel of the PIEZO1 trimer, likely through lateral tension on the membrane that deforms the PIEZO1 dome (inferred from the mouse homolog). In mouse osteocytes, Piezo1 is colocalized with Gja1 (Connexin 43) at the plasma membrane and the colocalization increases as Piezo1 is stimulated by force (Zeng et al. 2022).
