Active LTK appears to interact with SHC proteins through tyrosine 862 in an NPXY motif near the receptor C-terminus (Ueno et al, 1995; Ueno et al, 1996; Ueno et al, 1997). In the context of a chimeric EGFR-LTK receptor, the p46, p52 and p66 isoforms of SHC1 were co-precipitated with the receptor, GRB2 and SOS1 (Ueno et al, 1995). Signaling through the SHC1:GRB2:SOS pathway promotes activation of the MAPK signaling pathway, including phosphorylation of MAPK1 and MAPK3, and contributes to mitogenic cues (Ueno et al, 1995; Yamada et al, 2008; Roll and Reuther, 2012).
