The transcription factor Kruppel-like factor 4 (KLF4) directly binds to the GC-rich/E-box region in the CDH1 gene promoter to activate CDH1 gene transcription (Yori et al. 2010, Koopmansch et al. 2013). The KLF4 binding site in the CDH1 gene promoter partially overlaps with the ZEB2 binding site. Binding of ZEB2, a transcriptional repressor of the CDH1 gene, interferes with KLF4 binding (Koopmansch et al. 2013). Transcription factors PBX1 and MEIS2 may form a complex with KLF4 and facilitate KLF4 binding to the CDH1 gene promoter (Bjerke et al. 2011). Direct binding of KLF4 to the CDH1 gene promoter is evolutionarily conserved in mouse, where KLF4-mediated upregulation of CDH1 occurs downstream of the nuclear factor I-C (NFIC) during dentogenesis (Lee et al. 2014). KLF4 may bind to the CDH1 gene promoter cooperatively with the homeobox transcription factor MSX2 (Lin et al. 2018). Depending on the cellular context, KLF4 binding to the CDH1 gene promoter may have a repressive effect on the CDH1 gene transcription (Lai et al. 2012).
