A recombinant human TWIST1 was shown to repress transcription from the human CDH1 gene promoter fused to the luciferase gene in a luciferase reporter assay (Vesuna et al. 2008, Yang et al. 2012), possibly in cooperation with the histone methyltransferase SET8 (Yang et al. 2012). By immunoblotting, protein levels of TWIST1 and CDH1 were shown to be inversely correlated in breast cancer cell lines (Vesuna et al. 2008, Yang et al. 2012), with reduction in CDH1 protein levels more prominent when SET8 was overexpressed alongside TWIST1 (Yang et al. 2012).
TRIM29 (also known as ATDC), an E3 ubiquitin ligase implicated in innate immunity (Xing et al. 2016), has been reported as a TWIST1 transcriptional target which, through a negative feedback loop, downregulates TWIST1, leading to increased CDH1 expression (Ai et al. 2014).
