After initial activation of NKX2-5 and GATA4 by MESP1, GATA4 binds the promoter of the NKX2-5 gene and activates transcription (inferred from mouse homologs). SMAD1:SMAD4 complexes that are apparently activated by BMP signaling also bind the NKX2-5 promoter, recruit GATA4, and synergistically enhance transcription (inferred from mouse homologs). TBX20 binds the cardiac enhancer of the NKX2-5 promoter and activates transcription (inferred from mouse homologs)
