SLC26A4 (Pendrin, PDS) can mediate the efflux of iodide (I-) from the apical membranes of thyroid cells in exchange for a chloride (Cl-) ion, a function demonstrated in human HEK293 and monkey COS-7 cells (Yoshida et al., 2004; Dossena et al. 2006). SLC26A4 is highly expressed in the adult thyroid and its activity is necessary for production of thyroid hormone (Yoshida et al., 2004; Honda & Griffith, 2022). SLC26A4 is also expressed in the inner ear (Dossena et al. 2006; Honda & Griffith, 2022). Mutations in this gene are associated with Pendred syndrome, an autosomal-recessive disease which is one of the most common forms of syndromic deafness and associated goiter (Dossena et al. 2006; reviewed in Honda & Griffith, 2022).
