  • FUNCTION The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion (PubMed:14627637, PubMed:19029296, PubMed:19244346, PubMed:20717956, PubMed:21248079, PubMed:22129970, PubMed:26032412). RAB11B plays a role in endocytic recycling, regulating apical recycling of several transmembrane proteins including cystic fibrosis transmembrane conductance regulator/CFTR, epithelial sodium channel/ENaC, potassium voltage-gated channel, and voltage-dependent L-type calcium channel. May also regulate constitutive and regulated secretion, like insulin granule exocytosis. Required for melanosome transport and release from melanocytes. Also regulates V-ATPase intracellular transport in response to extracellular acidosis (PubMed:14627637, PubMed:19029296, PubMed:19244346, PubMed:20717956, PubMed:21248079, PubMed:22129970). Promotes Rabin8/RAB3IP preciliary vesicular trafficking to mother centriole by forming a ciliary targeting complex containing Rab11, ASAP1, Rabin8/RAB3IP, RAB11FIP3 and ARF4, thereby regulating ciliogenesis initiation (PubMed:25673879). On the contrary, upon LPAR1 receptor signaling pathway activation, interaction with phosphorylated WDR44 prevents Rab11-RAB3IP-RAB11FIP3 complex formation and cilia growth (PubMed:31204173). Also interacts with RABL3 to promote ciliary vesicle formation (PubMed:36052645).CATALYTIC ACTIVITY GTP + H2O = GDP + phosphate + H(+)COFACTOR Regulated by guanine nucleotide exchange factors (GEFs) which promote the exchange of bound GDP for free GTP (Probable). Regulated by GTPase activating proteins (GAPs) which increase the GTP hydrolysis activity (Probable). Inhibited by GDP dissociation inhibitors (GDIs) which prevent Rab-GDP dissociation (Probable).SUBUNIT Interacts with KCNMA1 (By similarity). Interacts with RAB11FIP1, RAB11FIP2, RAB11FIP3 and RAB11FIP4 (PubMed:11495908). May interact with TBC1D14 (PubMed:22613832). Interacts with ATP6V1E1 (PubMed:20717956). Interacts with PI4KB (PubMed:23572552). Interacts (GDP-bound form) with ZFYVE27 (PubMed:21976701). Interacts (GDP-bound form) with KIF5A in a ZFYVE27-dependent manner (PubMed:21976701). Interacts with RELCH (By similarity). Interacts (in GTP-bound form) with TBC1D8B (via domain Rab-GAP TBC) (PubMed:30661770). Forms a complex containing RAB11B, ASAP1, Rabin8/RAB3IP, RAP11FIP3 and ARF4 (PubMed:25673879). Interacts with WDR44 (PubMed:31204173).INTERACTION Recruited to phagosomes containing S.aureus. Colocalizes with RAB11AFIP1 on punctate vesicles (PubMed:26032412).ALTERNATIVE PRODUCTS Up-regulated by extracellular acidosis and down-regulated by alkalosis (at protein level).DOMAIN Switch I, switch II and the interswitch regions are characteristic of Rab GTPases and mediate the interactions with Rab downstream effectors. The switch regions undergo conformational changes upon nucleotide binding which drives interaction with specific sets of effector proteins, with most effectors only binding to GTP-bound Rab.PTM Citrullinated by PADI4.PTM (Microbial infection) Glycosylated on arginine residues by S.typhimurium protein Ssek3.DISEASE The disease is caused by variants affecting the gene represented in this entry.SIMILARITY Belongs to the small GTPase superfamily. Rab family.
