text TBX21 (Jenner et al. 2009, Kanhere et al. 2012, and inferred from mouse homologs in Djuretic et al. 2007) and RUNX3 (inferred from mouse homologs in Djuretic et al. 2007, Naoe et al. 2007) bind the IL4 gene and repress its expression (Kitamura et al. 2007, Jenner et al. 2009, and inferred from mouse homologs in Djuretic et al. 2007, Zhuang et al. 2009, Zhu et al. 2012). TBX21 binds a silencer located about 2 kb downstream of the IL4 coding region named HS IV (Kanhere et al. 2012). In mouse Th1 cells, both T-bet and Runx3 bind to this Il4 silencer (Djuretic et al. 2007). IL4 is an activator of Th2 differentiation, therefore T-Bet and RUNX3 act to restrict differentiation to the Th1 cell type.
