text MITF-M binds to the GXLYT2 gene to repress expression in melanoma cells as assessed by ChIP-seq, CRISPR and si/shRNA-mediated knockdown and CUT-and-RUN DNA binding analysis (Laurette et al, 2015; Webster et al, 2014; Dilshat et al, 2021). GXYLT2 encodes a glycosyltransferase that extends glycosylation groups of NOTCH proteins (Sethi et al, 2010). GXYLT2 has been implicated in the regulation of cell growth and migration in cancer through its impact on Notch signaling and may also play a role in epithelial-to-mesenchymal transition in gastric cancer (Cui et al, 2019; Song et al, 2023; Zhao and Cai, 2025), consistent with a role for MITF-M in regulating these processes (reviewed in Goding and Arnheiter, 2019; Rambow et al, 2019).
