text STAT1 dimer (Signal Transducer and Activator of Transcription 1) regulates CD274 (PD-L1) transcription primarily in response to interferon-gamma (IFN-?) signaling. Upon IFN-? stimulation, STAT1 becomes phosphorylated and forms dimers that translocate to the nucleus. There, STAT1 binds to gamma-activated sequence (GAS) elements within the PD-L1 promoter. This binding recruits transcriptional co-activators and the transcriptional machinery, enhancing PD-L1 gene transcription. The upregulation of PD-L1 by STAT1 helps tumor cells evade immune detection by inhibiting T cell activity (Padmanabhan et al., 2022, Liu et al., 2020)
