PSMF1 (PI31) is a proteasome inhibitor that binds to the 20S core particle (20S CP) (McCutchen-Maloney et al. 2000: human PSMF1 and bovine 20S CP were used; Hsu et al. 2023: human PSMF1 and bovine 20S CP were used) and inhibits its catalytic activity likely by inserting into the central cavity and preventing substrates from entering the cavity (Hsu et al. 2023: human PSMF1 and bovine 20S CP were used; Wang et al. 2024: human PSMF1 and bovine 20S CP were used), as well as by interfering with the association between the 20S CP and the activating regulatory particles, 19S RP (McCutchen et al. 2000: human PSMF1 and bovine 20S CP and 19S RP were used) and PA28 (Zaiss et al. 1999: mouse Psmf1, 20S CP, and mouse PA28alpha and PA28beta were used). PSMF1 has also been reported to bind and inhibit the 20S immunoproteasome core particle (20S iCP), but to a lesser extent than the canonical 20S CP (Wang et al. 2024). PSMF1 has been reported to interfere with the assembly of the 20S iCP (Zaiss et al. 2002). |
