text Expression of the AGPAT2 gene, encoding 1-acyl-sn-glycerol-3-phosphate acyltransferase beta that localizes to the endoplasmic reticulum membrane and is responsible for conversion of lysophosphatidic acid (LPA) into phosphatidic acid (PA) is positively regulated by PPARG:RXRA heterodimers and KMT2C (MLL3)/KMT2D (MLL4) histone methyltransferases. Expression of AGPAT2 mRNA is increased in fat tissue of human subjects treated with PPARG agonist pioglitazone, and correlates with increased mRNA levels of other PPARG responsive genes, such as SCD (Yao-Borengasser et al. 2008). Expression of the AGPAT2 gene is severely reduced in differentiating mouse brown preadipocytes expressing histone H3.3 K4M mutant or Kmt2c (Mll3) and Kmt2d (Mll4) with deleted SET domains (Jang et al. 2019: RNA-seq, supplementary information). Based on a mouse model of hepatic steatosis, expression of the AGPAT2 gene is directly stimulated by PPARG2 and the MLL4 complex (Kim et al. 2016).
