text Expression of the PEX11A gene, encoding Peroxisomal membrane protein 11A, is positively regulated in fat tissue by the PPARG:RXRA heterodimer and KMT2C (MLL3)/KMT2D (MLL4) histone methyltransferase complexes. Mouse Pex11a gene is clustered together with the Plin1 gene on chromosome 7. A peroxisome proliferator response element (PPRE) is found in between the Pex11a gene and the downstream Plin1 gene, at ~8 kb downstream of Pex11a transcription start site (TSS), and ~2 kb upstream of the Plin1 TSS. The Pparg:Rxra heterodimer specifically binds this PPRE to stimulate transcription of both Pex11a and Plin1 (Shimizu et al. 2004, Shimizu et al. 2006). The arrangement of the PEX11A and PLIN1 genes and their shared regulatory elements are conserved in rats and humans (Shimizu et al. 2006). Expression of the Pex11a gene is severely reduced in differentiating mouse brown preadipocytes expressing histone H3.3 K4M mutant or Kmt2c (Mll3) and Kmt2d (Mll4) with deleted SET domains (Jang et al. 2019: RNA-seq, supplementary information).
