Direct repression of CDH1 (E-cadherin) gene transcription by ZEB2 (Zinc finger E-box-binding homeobox 2, also known as SIP1) has been reported in multiple studies. A recombinant mouse Zeb2 was shown to repress transcription from the canine CDH1 gene promoter by luciferase reporter assays (Comijn et al. 2001, van Grunsven et al. 2003, Nam et al. 2014) and to also repress the endogenous canine CDH1 gene transcription in Madin-Darby canine kidney cells; (Grunsven et al. 2003). A recombinant mouse Zeb2 was shown to repress transcription from the human CDH1 gene promoter in a luciferase reporter assay as well as from the endogenous human CDH1 gene promoter (Vandewalle et al. 2005). A recombinant human ZEB2 represses transcription from the human CDH1 promoter by a luciferase reporter assay (Ghoumid et al. 2013). Missense mutations in ZEB2 from Mowat-Wilson syndrome patients abrogate ZEB2-mediated repression of CDH1 gene transcription by impairing binding of ZEB2 to the CDH1 gene promoter (Ghoumid et al. 2013).

Sumoylation of ZEB2 relieves ZEB2-mediated repression of CDH1 gene expression (Long et al. 2005). CdGAP (ARHGAP31) may act as a transcriptional co-repressor of ZEB2 in repression of CDH1 gene transcription (He et al. 2017, Ben Djoudi Ouadda et al. 2018).
