TOM40:TOM70 complex translocates proteins from the cytosol to the mitochondrial intermembrane space
The TOM40:TOM70 Complex transports precursor proteins from the cytosol, across the outer membrane of the mitochondrion, and into the intermembrane space from where they may be targeted to all locations within the mitochondrion. TOM40, TOM22, TOM5, TOM6, and TOM7 form the general import pore across the membrane (Dekker et al. 1998). TOM20 and TOM22 interact with presequences on mitochondrial precursors (Haucke et al. 1995, Brix et al. 1997, Meisinger et al. 2001, Yamano et al. 2008). TOM70 interacts with hydrophobic sequences in mature internal regions of mitochondrial proteins (Schlossman et al. 1994, Brix et al. 1997, Yamamoto et al. 2009).
Substrates of the TOM40:TOM70 complex include ATP1 (Yamamoto et al. 2009), ATP2 (Yamamoto et al. 2009), ATP9 (Hines et al. 1990), TOM40 (Yamamoto et al. 2009), SSC1 (Yamamoto et al. 2009), CIT1 (Yamamoto et al. 2009), ACO1 (Yamamoto et al. 2009), IDH1 (Yamamoto et al. 2009), BCS1 (Stan et al. 2003), CYT1 (Diekert et al. 2001), TIM54 (Kurz et al. 1999), TIM22 (Kurz et al. 1999), AAC (Sollner et al. 1990, Kurz et al. 1990, Kubrich et al. 1998, Endres et al. 1999, Truscott et al. 2002, Vasiljev et al. 2004), HSP60 (Yamano et al. 2008), CYB2 (Krayl et al. 2007). Some proteins such as Aco1, Atp1, Cit1, Idh1, and Atp2 contain both presequences that interact with TOM20 and mature regions that interact with TOM70 (Yamamoto et al. 2009).
