Programmed cell death 6-interacting protein (PDCD6IP, ALIX) is an accessory protein interacting with components of both ESCRT-I and ESCRT-III complexes of the ESCRT pathway (Martin-Serrano et al., 2003). DENV NS3 binds PDCD6IP, an interaction occurring during late infection or at viral egress that is critical for viral propagation, as its downregulation significantly reduces viral titer. The mechanism of this interaction is unclear (Khadka et al., 2011; Thepparit et al., 2019; reviewed in Rivera-Cuevas & Carruthers, 2023). Increased PDCD6IP expression has been found already in early phases of DENV infection (Pattanakitsakul et al., 2010). Data from Yellow Fever Virus (YFV) point to a role of this reaction in virion egress (Carpp et al., 2011).
