In studies involving human cell lines, DENV NS1 protein was shown to bind directly to matrix metalloproteinase-9 (MMP9) and zona occludens protein 1 (ZO-1). This interaction contributes to the disruption of junctions between human endothelial cells and leads to vascular leakage in mouse models, a potential factor in Dengue hemorrhagic fever. Additionally, NS1 was found to enhance MMP9 expression in human macrophages. These results were obtained from experiments with the Dengue serotype-2 (DENV-2) NGC strain (Pan et al., 2021).
