DENV NS3 and NS5 interact (Takahashi et al., 2012). The polymerase activity of NS5 probably requires the helicase and the NTPase activity of NS3 to replicate the genome. Also, 5′-end capping of nascent RNA involves coordinated activities of NS3 (RNA 5′-triphosphatase) and the NS5 N-terminal SAM-dependent methyltransferase (N7- and 2′-O methylation) (Johansson et al., 2001 using DENV-2 strain TSV01).
