A portion of nucleus-localized Dengue virus (DENV) NS5 protein is exported to cytosolic regions. This export process, demonstrated in monkey (Vero) and human (HEK293T, 293) cells using DENV-2 strains (New Guinea C and Townsville), requires binding to XPO1 (CRM1) through a nuclear export signal (NES) at residues 327-343 (2818-2834 in the polyprotein) of the NS5 protein (Rawlinson et al., 2009). We display subsequent association with the ER membrane.
