The NS3 peptidase domain of the folded Dengue virus polyprotein, together with the upstream NS2B cofactor, can process the polyprotein "in cis" through autolytic cleavage. This has been demonstrated by cis-processing of DENV-2 polyprotein constructs at sites including NS2A/NS2B and NS2B/NS3 in vitro (Preugschat et al., 1990), and through cis autolytic cleavage at the NS2B/NS3 junction by recombinant Dengue virus (DENV-2, DENV-4) NS2B-NS3 proteins (Bera et al., 2007). Such NS3-mediated processing of DENV-2 polyprotein constructs results in the production of the C-prM-E-NS1-NS2A fragment, NS2B, and the NS345 fragment (Preugschat & Strauss, 1991). The ability of recombinant DENV-2 NS2B-NS3 to perform cis autolytic cleavage at the NS2B/NS3 junction contributes to forming NS2B and the NS3-containing fragment (Bera et al., 2007), and this enzyme can also cleave peptide substrates representing the NS2A/NS2B and NS2B/NS3 junctions (Shiryaev et al., 2007).
