STAT3 (Signal Transducer and Activator of Transcription 3) regulates CD274 (PD-L1) transcription in response to various cytokines and growth factors, including IL-6. Upon activation, STAT3 becomes phosphorylated, dimerizes, and translocates to the nucleus, where it binds to specific STAT3-responsive elements in the PD-L1 promoter region. This binding recruits transcriptional co-activators and the transcriptional machinery, leading to increased PD-L1 gene transcription (Marzec et al., 2008, Song et al., 2018, Tong et al., 2020).
